NM_006231.4(POLE):c.542A>T (p.Asp181Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D181V variant (also known as c.542A>T), located in coding exon 6 of the POLE gene, results from an A to T substitution at nucleotide position 542. The aspartic acid at codon 181 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
