It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PPM1K are known to be pathogenic (PMID: 23086801, 40047138). This variant is present in population databases (rs748117308, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with PPM1K-related conditions. For these reasons, this variant has been classified as Pathogenic.
