NM_006231.4(POLE):c.3623G>A (p.Ser1208Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1208N variant (also known as c.3623G>A), located in coding exon 30 of the POLE gene, results from a G to A substitution at nucleotide position 3623. The serine at codon 1208 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
