NM_006231.4(POLE):c.3539A>G (p.Gln1180Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1180R variant (also known as c.3539A>G), located in coding exon 29 of the POLE gene, results from an A to G substitution at nucleotide position 3539. The glutamine at codon 1180 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
