Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.1855T>C (p.Cys619Arg), citing Ambry Variant Classification Scheme 2023: The p.C619R variant (also known as c.1855T>C), located in coding exon 17 of the POLE gene, results from a T to C substitution at nucleotide position 1855. The cysteine at codon 619 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
