Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.1642G>A (p.Glu548Lys), citing Ambry Variant Classification Scheme 2023: The p.E548K variant (also known as c.1642G>A), located in coding exon 15 of the POLE gene, results from a G to A substitution at nucleotide position 1642. The glutamic acid at codon 548 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
