This variant lies in the KCNT1 gene (transcript NM_020822.3) at 6 bases into the intron immediately after coding-DNA position 1619, where T is replaced by A. Submitter rationale: This variant has not been reported in the literature in individuals with KCNT1-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change falls in intron 16 of the KCNT1 gene. It does not directly change the encoded amino acid sequence of the KCNT1 protein, but it affects a nucleotide within the consensus splice site of the intron. Nucleotide substitutions within the consensus splice site are relatively common causes of aberrant splicing (PMID: 17576681, 9536098).
