Uncertain significance for BAP1-related tumor predisposition syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004656.4(BAP1):c.659+2dup, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 8 of the BAP1 gene. It does not directly change the encoded amino acid sequence of the BAP1 protein. RNA analysis indicates that this variant induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BAP1-related conditions. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Studies have shown that this variant results in activation of a cryptic splice site, and produces a non-functional protein and/or introduces a premature termination codon (internal data). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
