Uncertain significance for Combined oxidative phosphorylation defect type 14 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006567.5(FARS2):c.563A>G (p.Tyr188Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with cysteine at codon 188 of the FARS2 protein (p.Tyr188Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with a FARS2-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, this variant has uncertain impact on FARS2 function. The available evidence is currently insufficient to determine its role in disease.
