This variant lies in the CRPPA gene (transcript NM_001101426.4) at coding-DNA position 356, where T is replaced by A; at the protein level this means replaces leucine at residue 119 with glutamine — a missense variant. Submitter rationale: This sequence change replaces leucine with glutamine at codon 119 of the ISPD protein (p.Leu119Gln). The leucine residue is moderately conserved and there is a moderate physicochemical difference between leucine and glutamine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with ISPD-related disease.
