Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000336.3(SCNN1B):c.1850_1851insT (p.Asn619fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Asn619Glnfs*3) in the SCNN1B gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 22 amino acid(s) of the SCNN1B protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with autosomal dominant Liddle syndrome (PMID: 25378078, 27896928). In at least one individual the variant was observed to be de novo. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
