Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_012472.6(DNAAF11):c.1096A>G (p.Lys366Glu), citing Ambry Variant Classification Scheme 2023: The p.K366E variant (also known as c.1096A>G), located in coding exon 10 of the LRRC6 gene, results from an A to G substitution at nucleotide position 1096. The lysine at codon 366 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
