This variant lies in the TRPM4 gene (transcript NM_017636.4) at coding-DNA position 1263 through 22 bases into the intron immediately after coding-DNA position 1263, deleting this region. Submitter rationale: This variant results in the deletion of part of exon 10 (c.1263_1263+22del) of the TRPM4 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in TRPM4 cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TRPM4-related conditions.
