Pathogenic for Familial melanoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000077.5(CDKN2A):c.202_203delinsTA (p.Ala68Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CDKN2A gene (transcript NM_000077.5) at coding-DNA position 202 through coding-DNA position 203, replacing the reference sequence with TA; at the protein level this means converts the codon for alanine at residue 68 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The CDKN2A gene encodes two different proteins, p16INK4a and p14ARF, which are translated from alternative transcripts with different open reading frames. Both transcripts have been analyzed. We report either the variant with the higher classification or default to the CDKN2A (p16INK4a) variant. This report therefore includes the details for the CDKN2A (p16INK4a) variant. This sequence change creates a premature translational stop signal (p.Ala68*) in the CDKN2A (p16INK4a) gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CDKN2A (p16INK4a) are known to be pathogenic (PMID: 15146471, 16905682). Information on the frequency of this variant in the gnomAD database is not available, as this variant may be reported differently in the database. This variant has not been reported in the literature in individuals affected with CDKN2A (p16INK4a)-related conditions.
