Pathogenic for Short-rib thoracic dysplasia 6 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001199397.3(NEK1):c.1507dup (p.Ile503fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ile503Asnfs*2) in the NEK1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NEK1 are known to be pathogenic (PMID: 22499340, 29068549). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NEK1-related conditions. For these reasons, this variant has been classified as Pathogenic.
