Uncertain significance for MEGF10-related myopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001256545.2(MEGF10):c.1044G>T (p.Glu348Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with aspartic acid at codon 348 of the MEGF10 protein (p.Glu348Asp). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and aspartic acid. This variant is present in population databases (rs370219469, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with MEGF10-related conditions. ClinVar contains an entry for this variant (Variation ID: 472732). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The aspartic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
