NM_004656.4(BAP1):c.356C>G (p.Thr119Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T119S variant (also known as c.356C>G), located in coding exon 5 of the BAP1 gene, results from a C to G substitution at nucleotide position 356. The threonine at codon 119 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
