Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_004656.4(BAP1):c.1806G>C (p.Glu602Asp), citing ACMG Guidelines, 2015: This missense variant replaces glutamic acid with aspartic acid at codon 602 of the BAP1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in one individual affected with uveal melanoma in the literature (PMID: 23171164). This variant has been identified in 1/31408 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
