This variant lies in the LAMB2 gene (transcript NM_002292.4) at coding-DNA position 3547, where A is replaced by G; at the protein level this means replaces isoleucine at residue 1183 with valine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with valine at codon 1183 of the LAMB2 protein (p.Ile1183Val). The isoleucine residue is weakly conserved and there is a small physicochemical difference between isoleucine and valine. This variant is present in population databases (rs370877359, ExAC 0.002%) but has not been reported in the literature in individuals with a LAMB2-related disease.
