This variant lies in the SLC2A1 gene (transcript NM_006516.4) at coding-DNA position 1358, deleting one base; at the protein level this means shifts the reading frame starting at proline residue 453, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change is expected to alter the c-terminus of the SLC2A1 protein (p.Pro453Leufs*55). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 40 amino acid(s) of the SLC2A1 protein and extend the protein by 14 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SLC2A1-related conditions. This variant disrupts a region of the SLC2A1 protein in which other variant(s) (p.Arg458Trp) have been determined to be pathogenic (PMID: 23280796, 23340081, 28116237). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
