This variant lies in the NBN gene (transcript NM_002485.5) at coding-DNA position 896, where G is replaced by C; at the protein level this means replaces arginine at residue 299 with threonine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with threonine, which is neutral and polar, at codon 299 of the NBN protein (p.Arg299Thr). This variant also falls at the last nucleotide of exon 7, which is part of the consensus splice site for this exon. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NBN-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
