Uncertain significance for Galactosylceramide beta-galactosidase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000153.4(GALC):c.1249T>A (p.Phe417Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 417 of the GALC protein (p.Phe417Ile). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GALC-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
