Uncertain significance for Hereditary diffuse gastric adenocarcinoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004360.5(CDH1):c.387+1111C>T, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CDH1 gene (transcript NM_004360.5) at 1111 bases into the intron immediately after coding-DNA position 387, where C is replaced by T. Submitter rationale: This sequence change falls in intron 3 of the CDH1 gene. It does not directly change the encoded amino acid sequence of the CDH1 protein. RNA analysis indicates that this variant induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CDH1-related conditions. Studies have shown that this variant results in activation of a cryptic splice site, and produces a non-functional protein and/or introduces a premature termination codon (internal data). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
