This variant lies in the FLNC gene (transcript NM_001458.5) at coding-DNA position 4888, where G is replaced by T; at the protein level this means replaces alanine at residue 1630 with serine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with FLNC-related disease. This sequence change replaces alanine with serine at codon 1630 of the FLNC protein (p.Ala1630Ser). The alanine residue is highly conserved and there is a moderate physicochemical difference between alanine and serine.
