Pathogenic for DICER1-related tumor predisposition — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_177438.3(DICER1):c.4050+2T>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 21 of the DICER1 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individuals with DICER1-related conditions (PMID: 21882293, 25022261). Studies have shown that disruption of this splice site results in activation of a cryptic splice site, and produces a non-functional protein and/or introduces a premature termination codon (internal data). For these reasons, this variant has been classified as Pathogenic.
