NM_015192.4(PLCB1):c.3606G>C (p.Glu1202Asp) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1202D variant (also known as c.3606G>C), located in coding exon 32 of the PLCB1 gene, results from a G to C substitution at nucleotide position 3606. The glutamic acid at codon 1202 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
