Submitter rationale: This sequence change affects the initiator methionine of the NPC1 mRNA. The next in-frame methionine is located at codon 156. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NPC1-related conditions. This variant disrupts a region of the NPC1 protein in which other variant(s) (p.Gln92Arg ) have been determined to be pathogenic (PMID: 11349231, 11479732, 28105569, 31139477).
