NM_000071.3(CBS):c.1205T>C (p.Leu402Pro) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L402P variant (also known as c.1205T>C), located in coding exon 11 of the CBS gene, results from a T to C substitution at nucleotide position 1205. The leucine at codon 402 is replaced by proline, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
