Uncertain significance for Neutropenia, severe congenital, 2, autosomal dominant — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005263.5(GFI1):c.357del (p.Phe119fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Phe119Leufs*79) in the GFI1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in GFI1 cause disease. This variant is present in population databases (rs770845131, gnomAD 0.001%). This variant has not been reported in the literature in individuals affected with GFI1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
