Uncertain significance for KBG syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_013275.6(ANKRD11):c.406A>C (p.Thr136Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 136 of the ANKRD11 protein (p.Thr136Pro). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ANKRD11-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt ANKRD11 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
