This variant lies in the SYNE1 gene (transcript NM_182961.4) at coding-DNA position 242, where G is replaced by A; at the protein level this means replaces arginine at residue 81 with histidine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
