Pathogenic for Congenital myasthenic syndrome 4A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000080.4(CHRNE):c.1032+1G>T, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CHRNE gene (transcript NM_000080.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1032, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 9 of the CHRNE gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individuals with congenital myasthenic syndrome (PMID: 11408331, 37946251). This variant is also known as IVS9+1G>T. Studies have shown that disruption of this splice site results in skipping of exon 9, and produces a non-functional protein and/or introduces a premature termination codon (PMID: 11408331). For these reasons, this variant has been classified as Pathogenic.
