Uncertain significance for Emery-Dreifuss muscular dystrophy 4, autosomal dominant; Autosomal recessive ataxia, Beauce type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_182961.4(SYNE1):c.21602G>A (p.Cys7201Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with tyrosine at codon 7130 of the SYNE1 protein (p.Cys7130Tyr). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and tyrosine. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a SYNE1-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies. In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance.
