Pathogenic for Charcot-Marie-Tooth Neuropathy X — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000166.6(GJB1):c.253T>C (p.Ser85Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 85 of the GJB1 protein (p.Ser85Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of Charcot-Marie-Tooth disease (external communication). In at least one individual the variant was observed to be de novo. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt GJB1 protein function with a positive predictive value of 95%. This variant disrupts the p.Ser85 amino acid residue in GJB1. Other variant(s) that disrupt this residue have been observed in individuals with GJB1-related conditions (PMID: 9099841), which suggests that this may be a clinically significant amino acid residue. For these reasons, this variant has been classified as Pathogenic.
