This variant lies in the TMTC3 gene (transcript NM_181783.4) at coding-DNA position 212, where G is replaced by A; at the protein level this means replaces arginine at residue 71 with histidine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 71 of the TMTC3 protein (p.Arg71His). This variant is present in population databases (rs770896677, gnomAD 0.01%). This missense change has been observed in individual(s) with clinical features of TMTC3-related conditions (PMID: 28973161). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. For these reasons, this variant has been classified as Pathogenic.
