Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005654.6(NR2F1):c.437G>T (p.Cys146Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with phenylalanine, which is neutral and non-polar, at codon 146 of the NR2F1 protein (p.Cys146Phe). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NR2F1-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt NR2F1 protein function with a positive predictive value of 95%. This variant disrupts the p.Cys146 amino acid residue in NR2F1. Other variant(s) that disrupt this residue have been observed in individuals with NR2F1-related conditions (PMID: 26986877, 35791240), which suggests that this may be a clinically significant amino acid residue.
