Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000352.6(ABCC8):c.4515C>A (p.Asp1505Glu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ABCC8 gene (transcript NM_000352.6) at coding-DNA position 4515, where C is replaced by A; at the protein level this means replaces aspartic acid at residue 1505 with glutamic acid — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with glutamic acid, which is acidic and polar, at codon 1505 of the ABCC8 protein (p.Asp1505Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal dominant ABCC8-related conditions (PMID: 20573158, 20685672). In at least one individual the variant was observed to be de novo. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt ABCC8 protein function with a positive predictive value of 80%. For these reasons, this variant has been classified as Pathogenic.
