This variant lies in the PURA gene (transcript NM_005859.5) at coding-DNA position 115, where G is replaced by T; at the protein level this means replaces glycine at residue 39 with cysteine — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with cysteine, which is neutral and slightly polar, at codon 39 of the PURA protein (p.Gly39Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of PURA syndrome (internal data). In at least one individual the variant was observed to be de novo. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on PURA protein function.
