Pathogenic for Spondyloepiphyseal dysplasia with congenital joint dislocations — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004273.5(CHST3):c.802G>T (p.Glu268Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Glu268*) in the CHST3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 212 amino acid(s) of the CHST3 protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with spondyloepiphyseal dysplasia (PMID: 26572954). It has also been observed to segregate with disease in related individuals. For these reasons, this variant has been classified as Pathogenic.
