Likely pathogenic for Citrullinemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_054012.4(ASS1):c.872A>C (p.Tyr291Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with serine, which is neutral and polar, at codon 291 of the ASS1 protein (p.Tyr291Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of citrullinemia (PMID: 18925679; internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt ASS1 protein function with a negative predictive value of 80%.
