Uncertain significance for Emery-Dreifuss muscular dystrophy 5, autosomal dominant — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_182914.3(SYNE2):c.13695_13707+2dup, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SYNE2 gene (transcript NM_182914.3) at coding-DNA position 13695 through the canonical splice donor site of the intron immediately after coding-DNA position 13707, duplicating this region. Submitter rationale: This sequence change falls in intron 72 of the SYNE2 gene. It does not directly change the encoded amino acid sequence of the SYNE2 protein. It affects a nucleotide within the consensus splice site. This variant is present in population databases (rs758985012, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with SYNE2-related conditions. ClinVar contains an entry for this variant (Variation ID: 470930). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Experimental studies and prediction algorithms are not available or were not evaluated, and the effect of this variant on mRNA splicing is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
