Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_004304.5(ALK):c.523A>G (p.Ser175Gly), citing Sema4 Curation Guidelines. This variant lies in the ALK gene (transcript NM_004304.5) at coding-DNA position 523, where A is replaced by G; at the protein level this means replaces serine at residue 175 with glycine — a missense variant. Submitter rationale: The ALK c.523A>G (p.S175G) variant has not been reported in the literature to our knowledge. It was observed in 1/113206 chromosomes of the Non-Finnish European subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 470889). In silico tools suggest the impact of the variant on protein function is benign, though these predictions have not been confirmed by functional studies. The overall evidence is insufficient to meet ACMG/AMP criteria for classifying it as benign or pathogenic. In summary, the clinical significance of this variant is currently uncertain.
