Submitter rationale: This sequence change replaces histidine, which is basic and polar, with aspartic acid, which is acidic and polar, at codon 446 of the MAN2B1 protein (p.His446Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with alpha mannosidosis (internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant.
