Uncertain significance for Dilated cardiomyopathy 1JJ — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001105206.3(LAMA4):c.4280del (p.Asn1427fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Asn1420Ilefs*18) in the LAMA4 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in LAMA4 cause disease. This variant is present in population databases (rs782604289, gnomAD 0.002%). This premature translational stop signal has been observed in individual(s) with dilated cardiomyopathy (PMID: 32880476). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
