This variant lies in the HCN4 gene (transcript NM_005477.3) at coding-DNA position 497, where C is replaced by T; at the protein level this means replaces proline at residue 166 with leucine — a missense variant. Submitter rationale: Variant summary: HCN4 c.497C>T (p.Pro166Leu) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 4.3e-05 in 93296 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.497C>T has been observed in at least one individual who died from Sudden Arrhythmic Death Syndrome, without strong evidence of causality (example: Nunn_2016). This report does not provide unequivocal conclusions about association of the variant with Sick Sinus Syndrome 2.
