Uncertain significance for Primary immunodeficiency with post-measles-mumps-rubella vaccine viral infection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005419.4(STAT2):c.2528A>T (p.Asp843Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with valine, which is neutral and non-polar, at codon 843 of the STAT2 protein (p.Asp843Val). This variant is present in population databases (rs766059622, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with STAT2-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
