NM_003072.5(SMARCA4):c.634G>A (p.Gly212Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the SMARCA4 gene (transcript NM_003072.5) at coding-DNA position 634, where G is replaced by A; at the protein level this means replaces glycine at residue 212 with arginine — a missense variant. Submitter rationale: The p.G212R variant (also known as c.634G>A), located in coding exon 3 of the SMARCA4 gene, results from a G to A substitution at nucleotide position 634. The glycine at codon 212 is replaced by arginine, an amino acid with dissimilar properties. This variant has been detected in multiple individuals with no reported features of Coffin-Siris syndrome (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the supporting evidence, the association of this alteration with rhabdoid tumor predisposition syndrome is unknown; however, the association of this alteration with Coffin-Siris syndrome is unlikely.
