Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032447.5(FBN3):c.167G>T (p.Gly56Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FBN3 gene (transcript NM_032447.5) at coding-DNA position 167, where G is replaced by T; at the protein level this means replaces glycine at residue 56 with valine — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 56 of the FBN3 protein (p.Gly56Val). This variant also falls at the last nucleotide of exon 2, which is part of the consensus splice site for this exon. This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with FBN3-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
