Uncertain significance for X-linked Emery-Dreifuss muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000117.3(EMD):c.128C>T (p.Thr43Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 43 of the EMD protein (p.Thr43Ile). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with EMD-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt EMD protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change does not substantially affect EMD function (PMID: 31185657). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
