Pathogenic for Familial adenomatous polyposis 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.10:g.(?_112827102)_(112844132_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 12-16 of the APC gene. The 5' boundary is likely confined to intron 11. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation. Deletions encompassing exons 12-16, also known as exons 11-15 in the literature, have been reported in individuals and families affected with familial adenomatous polyposis (PMID: 20223039, 16736293, 18487285). For these reasons, this variant has been classified as Pathogenic.
